Shedding Light Onto O-Glycosylation

For the past 12 years, my research group has been interested in the development of approaches to chemical glycosylation that leverage new and unusual forms of reactivity. Chief among our interests is the development of “mild” approaches to the activation of recalcitrant substrates such as thioglycosides. It is reasonable to expect stable substrates to be unreactive. However, irradiation, even with visible light, can provide the activation energy that might otherwise be supplied only with highly reactive activators. We have found that generation of electron-donor-acceptor (EDA) complexes between thioglycosides (and related substrates) and sulfonium ions serves as a prerequisite to effective photochemical activation and subsequent glycosylation.
