Review for Honors 298 Final Exam

 

Please review the following items for the final exam.  The final will not be comprehensive and will only cover the material from the last exam.  It will start with information presented on Recombinant DNA Technology and finish with Xenotransplantation.  The following is a list of topics to focus on:

 

1) Recombinant DNA technology.  What is a clone and how are clones made?  Embryo splitting and nuclear transfer.

 

2) Multistep process of cloning DNA fragments.  What are restriction enzymes?  What are vectors?  What is transformation and how this is done?  Antibiotic screening, other definitions.

 

3) What is a gene library?  What is a probe? 

 

4) What is DNA sequencing?  What is PCR? (know steps, cycle)

 

5) What is a Southern Blot?  How is this done?

 

6) What is DNA fingerprinting? What are minisatellites? (know details and applications)

 

7) Prenatal and presymptomatic analysis with DNA technology.

 

8) What is the Human Genome Project?  (benefits, goals, downside)

 

9) Microbial, plant and animal applications of genetic engineering.

 

10) Acceptance of Biotechnology survey.

 

11) Human gene therapy….basic principles (retrovirus), applications (ADA), problems.

 

12) Tissue engineering (liver, pancreas, skin, bladder) and ES cells.

 

13) Organ transplantation, HLA complex.

 

14) Xenotransplantation and problems.
